A LC-MS library for the identification of flavonoids was constructed from the Cactaceae family based on previous reports. A total of 24 flavonol derivatives were identified in the used parts of Opuntia ficus-indica var. saboten (prickly pear) and Hylocereus undatus (dragon fruit) using UPLC-DAD-QToF/MS and a constructed library. Among them, as new flavonol derivatives, isorhamnetin 3-methyl ether and kamepferol 3-O-galactoside (trifolin) were tentatively identified in prickly pear, and quercetin 3-O-robinobioside and isorhamnetin 3-O-rutinoside-7-O-rhamnoside were identified in dragon fruit (peels). In particular, isorhamnetin 3-O-rutinoside (narcissin) and isorhamnetin 3-O-robinobioside were confirmed as the major compounds in both samples. The flavonoid contents of prickly pear increased in the order of cladode (un-peeled)> whole fruit> cladode (peeled), and dragon fruit showed higher concentrations in the peels than pulps, suggesting that the by-products of these plants are more valuable as functional resources for various industries, such as foods, cosmetics, and pharmaceuticals.
